Packaging assessment

CryoTEM preparation and imaging allows to observe the specimen and its particles in a near-native hydrated state. Through this method, the inner content of the particles can be directly visualized and assessed, allowing a straight forward discrimination between particles rich in drug and empty ones.

Our semi-automated detection method using our in-house software solution presents a representative histogram (left) that effectively displays the liposome packaging. The corresponding image (right) simplifies data interpretation by overlaying detected and classified particles with clear blue/green/red outlines. This allows for a visual, user-friendly understanding of encapsulation efficiency. The percentage of encapsulation can be correlated to the efficiency of the drug release. This data is provided in the analytical report.

Lamellarity Assessment

Insights on the liposomes lamellarity is essential for predicting drug release kinetics. CryoTEM combined with image analysis from our software package allows to distinguish liposomes based on their lamellarity.
